    Antibodies for immunolabeling by light and electron microscopy : not for the faint hearted

    Reliable antibodies represent crucial tools in the arsenal of the cell biologist and using them to localize antigens for immunocytochemistry is one of their most important applications. However, antibody-antigen interactions are much more complex and unpredictable than suggested by the old 'lock and key' analogy, and the goal of trying to prove that an antibody is specific is far more difficult than is generally appreciated. Here, we discuss the problems associated with the very complicated issue of trying to establish that an antibody (and the results obtained with it) is specific for the immunolabeling approaches used in light or electron microscopy. We discuss the increasing awareness that significant numbers of commercial antibodies are often not up to the quality required. We provide guidelines for choosing and testing antibodies in immuno-EM. Finally, we describe how quantitative EM methods can be used to identify reproducible patterns of antibody labeling and also extract specific labeling distributions.Peer reviewe

    A Highly Flexible Bunch Compressor for the APS LEUTL FEL

    The LEUTL FEL at the APS has achieved gain at 530 nm with peak beam current \~100 A. In an effort to push to 120 nm and beyond, we have designed and are commissioning a bunch compressor to increase the peak current to 600 A or more. The bunch compressor uses a four-dipole chicane at an energy of 100 to 210 MeV. To provide options for control of emittance growth due to coherent synchrotron radiation (CSR), the chicane has variable R56. The central pair of dipoles is movable, accommodating variable R56 with less concern about emittance dilution from nonuniformity of the dipole field. The symmetry of the chicane is also variable via longitudinal motion of the final dipole, which is predicted to have an effect on emittance growth. Following the chicane, a three-screen emittance measurement system should permit resolution of the difference in emittance growth between various chicane configurations. A vertical bending magnet analysis line is present to permit imaging of correlations between transverse and energy coordinates. This paper reviews the features and expected performance of the chicane. A companion paper discusses the physics design in detail.     Integration effects of pylon geometry on a high-wing transport airplane

    An investigation was conducted in the Langley 16-Foot Transonic Tunnel to determine the installation effects of a series of pylons that had differing cross-sectional shapes on the pressure distributions and aerodynamic characteristics of a 1/24-scale high wing transport. The tests were conducted at Mach numbers at 0.70 and 0.80 at angles of attack from -3 degrees to 4 degrees with the pylons tested at various toe angles between 5 degrees inboard and 5 degrees outboard. Results of this study indicate that the installed drag was lowest for the pylons with a compression pylon type design which kept the flow under the wing in the pylon/wing junction comparable to the clean wing velocities

    A new image processing technique was applied to Viking Lander multispectral images. Spectral endmembers were defined that included soil, rock and shade. Mixtures of these endmembers were found to account for nearly all the spectral variance in a Viking Lander image

    Compositional variability of the Martian surface

    Spectral reflectance data from Viking Landers and Orbiters and from telescopic observations were analyzed with the objective of isolating compositional information about the Martian surface and assessing compositional variability. Two approaches were used to calibrate the data to reflectance to permit direct comparisons with laboratory reference spectra of well characterized materials. In Viking Lander multispectral images (six spectral bands) most of the spectral variation is caused by changes in lighting geometry within individual scenes, from scene to scene, and over time. Lighting variations are both wavelength independent and wavelength dependent. By calibrating lander image radiance values to reflectance using spectral mixture analysis, the possible range of compositions was assessed with reference to a collection of laboratory samples, also resampled to the lander spectral bands. All spectra from the lander images studied plot (in six-space) within a planar triangle having at the apexes the respective spectra of tan basaltic palagonite, gray basalt, and shale. Within this plane all lander spectra fit as mixtures of these three endmembers. Reference spectra that plot outside of the triangle are unable to account for the spectral variation observed in the images

    Internet Governance: the State of Play

    The Global Forum on Internet Governance held by the UNICT Task Force in New York on 25-26 March concluded that Internet governance issues were many and complex. The Secretary-General's Working Group on Internet Governance will have to map out and navigate this complex terrain as it makes recommendations to the World Summit on an Information Society in 2005. To assist in this process, the Forum recommended, in the words of the Deputy Secretary-General of the United Nations at the closing session, that a matrix be developed "of all issues of Internet governance addressed by multilateral institutions, including gaps and concerns, to assist the Secretary-General in moving forward the agenda on these issues." This paper takes up the Deputy Secretary-General's challenge. It is an analysis of the state of play in Internet governance in different forums, with a view to showing: (1) what issues are being addressed (2) by whom, (3) what are the types of consideration that these issues receive and (4) what issues are not adequately addressed
